摘要
目的探讨原花青素(OPC)对C57BL/6J小鼠血管内皮舒张功能的影响及物质基础。方法①将内皮功能紊乱的C57BL/6J小鼠分为对照组、0.1%w/w原花青素组、抗生素组、抗生素+0.1%w/w原花青素组,干预4周后观察小鼠胸主动脉舒张功能;②采用高效液相色谱法(HPLC)测定OPC(100mg/kg)灌胃后小鼠60min血浆中OPC以及其代谢物原儿茶酸(PCA)含量;③将内皮功能紊乱的C57BL/6J小鼠分为抗生素组、抗生素+0.1%w/wOPC、抗生素+0.1%w/wOPC+0.003%w/wPCA,干预4周后观察小鼠胸主动脉舒张功能。结果①OPC干预可提高小鼠动脉舒张功能,但该效应被抗生素阻断;②OPC灌胃小鼠60min后,HPLC法测得血浆PCA浓度为(10.6±1.8)μmol/L,但OPC未检出;③抗生素阻断OPC介导的小鼠动脉舒张功能可被其代谢物PCA部分恢复。结论原花青素可以改善血管内皮舒张功能,其物质基础可能与其代谢物PCA有关。
Objective To investigate the effect of oligomeric proanthocyanidins (OPC) on thoracic aorta endothelial function in C57BL/6J mice. Method (1)Forty male C57BL/6J mice were randomly divided into 4 groups: normal saline and 45% fat diet; normal saline and 0.1% w/w OPC in 45% fat diet; antibiotics cocktail and 45% fat diet group; antibiotics cocktail and 0.1% w/w OPC in 45% fat diet group. The thoracic aorta endothelial vasodilatation was measured after 4-week consumption. (2)Mice plasma were detected by high performance liquid chromatography (HPLC) after 60 min of OPC intragastric administration.(3)Another 30 male C57BL/6J mice were randomly divided into 3 groups:antibiotics cocktail group, antibiotics cocktail+OPC group, antibiotics cocktail+protocatechuic acid(PCA)+OPC group. The thoracic aorta endothelial vasodilatation was measured after 4-week consumption. Results (1)The endothelial vasodilatation of thoracic aorta in OPC group was significantly higher than that of the normal saline group.When antibiotics cocktail was used for treatment,the endothelial vasodilatation of thoracic aorta in OPC was blocked.(2)By using HPLC for detection, PCA concentration was (10.6±1.8)μmol/ L but OPC can not be detected in mice plasma. (3)The endothelial vasodilatation of thoracic aorta can be improved by PCA. Conclusion OPC improves the vascular endothelial function may via its metabolites PCA.
