摘要
目的探讨补中益气丸对乳腺癌化疗相关性疲劳的作用及其机制。方法将小鼠随机分为正常(NC)组和待处理组。将待处理组小鼠建成荷4T1乳腺癌小鼠模型,并将其随机分为肿瘤对照(TC)组、紫杉醇(PTX)组、补中益气丸(BZYQ)组、补中益气丸+紫杉醇(BZYQ+PTX)组。BZYQ1.5g/kg灌胃给药,每天1次;PTX10mg/kg腹腔注射,每2天1次。给药21d,观察小鼠体重、肿瘤体积、力竭游泳时间指标的变化,并检测肌肉SOD、MDA水平。结果与实验开始前比较,给药后第3周时NC组和BZYQ+PTX组小鼠的体重明显增加(P<0.05)。与TC组比较,给药第9d开始,BZYQ+PTX组小鼠的肿瘤体积明显变小(P<0.05),给药第18d时,PTX组小鼠的肿瘤体积变小(P<0.05);BZYQ+PTX组和PTX组两组小鼠的瘤重均变小(P<0.05~0.01)。与NC组比较,PTX组小鼠的平均力竭游泳时间则在给药后第1、2、3周均明显下降(P<0.05~0.01),PTX+BZYQ组小鼠的平均力竭游泳时间仅在给药第1周时下降(P<0.01),第2、3周时较PTX组小鼠明显延长(P<0.01)。与TC组比较,PTX组SOD值明显降低,而BZYQ+PTX组的SOD值较PTX组明显升高;PTX组MDA浓度明显升高,而BZYQ+PTX组的MDA浓度较PTX组明显降低。结论补中益气丸可能通过抑制氧化应激损伤发挥抗乳腺癌化疗相关性疲劳的作用。
Objective To evaluate the effect of Bu-Zhong-Yi-Qi pills in the alleviation of chemotherapy-induced fatigue in mice with breast cancer. Methods Mice injected with murine 4T1 mammary carcinoma ceils were randomly divided into five groups: negative control group (NC), tumor control group (TC), paclitaxel group (PTX), Bu-Zhong-Yi-Qi pills treatment group (BZYQ), and Bu-Zhong-Yi-Qi pills +paclitaxel group (BZYQ+PTX)group. The mice were then orally administered of the drugs for 21 days. The tumor volume, weight and the weight loaded swimming time were daily measured. After the last administration, all mice were sacrificed, and the blood and gastrocnemius muscle were collected and stored at -80℃ for the subsequent analysis of SOD and MDA. Results The body weight of mice in the negative control group and BZYQ+PTX group was significantly increased after 3 weeks of the treatment (P〈0.05).The tumor weight of the PTX group and BZYQ+ FIX group was apparently lower (P〈0.05) than the the tumor control group. The tumor volume in the BZYQ+PTX group was significandy reduced 9 days after the treatment(P〈0.05). The tumor volume in PTX group was significantly reduced after 18 days of treatment (P〈0.05). The weight loaded swimming time of the PTX group was apparently reduced in the first 3 weeks (P〈0.05). The weight loaded swimming time of the BZYQ+PTX group was apparently reduced in the first week (P〈0.01). Comparing with the PTX group, the weight loaded swimming time of the BZYQ group, NC group and TC group was significantly prolonged after 1 week of treatment(P〈0.01 ). The weight loaded swimming time of the other four groups of mice were significantly prolonged at the second and third weeks (P〈0.05). The level of SOD in the BZYQ+PTX group was significantly elevated,and the level of MDA in the BZYQ+PTX group was significantly lower than the FIX group (P〈0.05). Conclusion The Buzhong yiqi pills could alleviate chemotherapy-induced fatigue in mice bearing breast cancer cells.
