摘要
目的 :应用离体兔心Langendorff灌注模型 ,比较吲哒帕胺和维拉帕米对心肌再灌注损伤的保护作用 ,为临床应用提供实验依据。方法 :2 1只雄性家兔随机分为对照组、维拉帕米组和吲哒帕胺组 ;全心缺血 40min后再灌注 ,分别取再灌注 15、30、6 0和 36 0min左室壁心肌标本 ,做超微结构和肌动蛋白 (actin)免疫组化观察。结果 :对照组超微结构和actin改变明显 ,而维拉帕米组和吲哒帕胺组改变轻微。结论
Objective:The animal models of isolated myocardial postischemic reperfusion injury in rabbits using Langendorff apparatus were used to compare the protective effects of indapamide and verapamil on myocardial ischemia and reperfusion injury in order to give the experimental proof for clinic uses. Methods:Twenty-one male rabbits were randomly divided into three groups,( n =7 in each):control group, verapamil group, and indapamide group. Isolated hearts were reperfused after 40 min of global no-flow ischemia. Myocardial specimen of left ventricular wall was taken at 15, 30,60 and 360 min after postischemic reperfusion. Ultrastructure and actin were studied by using electron microscope and immunohistochemical methods respectively. Results:In the control group, marked ultrastructural and actin positive staining alterations were observed. In the verapamil group and the indapamide group, all morphological atlerations were mild. Conclusion:The results showed that the protective effects of indapamide on myocardial ischemia and reperfusion injury were better than that of verapamil.owed
