低浓度二甲基亚砜冰冻血小板的个性化研究

摘要 目的探讨血小板(PLT)冰冻保存时保护剂二甲基亚砜(DMSO)的最佳浓度。方法计算机随机选择2009年6月至2011年9月本血站机采室采集的120袋PLT作为研究对象。将不同质量浓度的DMSO作为保护剂加入PLT中,在-80℃条件下进行冰冻保存。检测冰冻前、后及不同离心条件下PLT计数、血浆中PLT第3因子(PF3)、PLT第4因子(PF4)、PLT体积分布宽度(PDW)及P-选择素(CD62P)的含量,并分析PLT的聚集、黏附、血块收缩功能的变化情况。结果质量浓度为2%与5%的DMSO作为保护剂时,对冰冻PLT的各项功能指标影响不大,差异无统计学意义(P〉0.05)。但1%的DMSO作为保护剂时,对PLT的各项功能指标影响较大。结论PLT为无核细胞,结构简单,可用2%的DMSO作为保护剂进行冷冻保存。 Objective To study the platelet (PLT) frozen when the best dose of dimethyl sulfoxide (DMSO) protectants, to reduce or avoid DMSO side effects. Methods From June 2009 to September 2011, a total of 120 bags of PLT were included in this study by computer randomly. PLT were added with different concentration of DMSO as a protective agent, and stored frozen in -80℃ conditions. The count of PLT, platelets third factor (PF3), platelet fourth factor (PF4), platelet volume distribution width (PDW) and P- selection (CD62P) were detected before and after frozen at different centrifugal condition. The change of PLT adhesion, gathered, clots systolic functions were analyzed, too. Results There was little effect on the frozen PLT function indicators when the protective agent was 2% DMSO or 5% DMSO, and the difference of each indicator was not statistically significant (P〉0.05). But there was a greater impact on the PLT function indicators, when the protective agent was 1% DMSO. Conclusions Because of non nuclear and simple structure of PLT, 2% DMSO could be used as cryopreservation protective agent.
