南方汉族人群HLA-DPA1和-DPB1基因多态性与鼻咽癌的关联研究

A Study on the Correlation of HLA-DPA1 and -DPB1 Polymorphisms With Nasopharyngeal Carcinoma in the Southern Chinese Han Population

摘要 目的探讨人类白细胞抗原(HLA)-DPA1和-DPB1基因多态性与中国南方汉族人群鼻咽癌的相关性。方法选择2012年2月至7月于广西梧州市红十字会医院及深圳市武警医院经病理学检查确诊为鼻咽癌(NPC)病变的南方汉族患者74例为研究对象,纳入研究组;计算机随机选择同期于本中心献血的南方汉族健康志愿捐者160例,纳入对照组(本研究遵循的程序符合本院人体试验委员会所制定的伦理学标准,得到该伦理会批准,分组征得受试对象本人的知情同意,并与之签订临床研究知情同意书)。采用测序分型方法对研究组和对照组的血液样本进行HLA-DPA1和-DPB1基因第2外显子双向测序,用Assign3.5SBT分析软件判定等位基因型。采用Wolf法进行z。检验并计算P值和相对危险度(RR),比较研究组和对照组HLA-DPA1和-DPB1等位基因频率的差异。结果研究组和对照组中,均检出4个DPA1等位基因,即DPA1*01:03,02:01,02:02和04:01。作为多态性丰富的HLA-DPB1位点,研究组中共检出14个等位基因,对照组中共检出18个等位基因。研究组与对照组的HLA-DPA1和-DPB1等位基因的基因检出率比较,差异均无统计学意义(χ2〈3.84,P〉0.05)。结论本文的结果初步显示中国南方汉族人群中HLA-DPA1和-DPB1基因多态性与NPC无相关性。 Objective To study the correlation of HLA-DPA1 and -DPB1 polymorphisms with nasopharyngeal carcinoma (NPC) in the southern Chinese Han population. Methods From February to July 2012,a total of 74 patients who diagnosed as NPC by pathological examination in Guangxi Wuzhou Red Cross Hospital and Shenzhen Armed Police Hospital were included in this study as study group. And during the same period, a total of 160 healthy donors who were selected by computer randomly in Shenzhen blood center were included in this study as control group (The study protocol was approved by the Ethical Review Board of Investigation in Human Being of our hospital). Both the study group and control group were from southern Chinese Han. All the samples were subjected to sequencing-based typing (PCR-SBT) at exon 2 of HLA-DPAI and -DPBI genes in both directions. The allelic frequencies of HLA-DPA1 and -DPB1 loci for the NPC patients were compared with control group by 3fz test, the P value and relative risk (RR) value were calculated according to the Wolf method. Results A total of 4 HLA-DPA1 alleles (DPA1* 01:03, 02:01, 02:02 and 04: 01) were identified in both study group and control group. As for the highly polymorphic HLA-DPB1 locus, 14 HLA-DPB1 alleles in the NPC patient group and 18 DPB1 alleles in the healthy controls were identified. The allele frequencies for both HLA-DPA1 and -DPB1 loci showed no significant differences between study group and control group (χ2〈3.84, P〈0.05). Conclusions The present study implied that the polymorphisms of HLA-DPA1 and -DPB1 showed no independent association with NPC in the southern Chinese Han individuals.
