摘要
[目的]研究石榴新品种"九州红"的组织培养及快繁技术。[方法]以"九州红"石榴当年生新梢茎段为外植体,以MS培养基为基本培养基,研究不同浓度激素配比对"九州红"芽分化、增殖及生根的影响。[结果]"九州红"最佳芽分化培养基为MS+0.8 mg/L6-BA+0.3 mg/L IAA+3.0%蔗糖+1.3%琼脂条,约40 d后可诱导出丛生芽,成芽率达61.2%;在MS+1.0 mg/L 6-BA+0.5 mg/L IAA+3.0%蔗糖+1.3%琼脂条培养基上丛生芽的增殖速度最快,增殖系数达3.62,且芽生长健壮;"九州红"的最佳生根培养基为1/2MS+0.6 mg/L IBA+0.1 mg/L NAA+3.0%蔗糖+1.3%琼脂条,此时生根率可达91.2%,平均每株生根达3.8条,且植株生长健壮,适于移栽。[结论]该研究为"九州红"石榴种苗的大规模生产提供了技术支持。
[ Objective ] The paper was to study the tissue culture and rapid propagation technology of new pomegranate variety ' Jiuzhou- hong'. [ Method ] The effects of different hormones combination and concentration on bud differentiation, proliferation and rooting culture were studied by using MS medium as the basic medium and using stems of new shoots grown in current year of ' Jiuzhouhong' as explants. [ Result] The best bud differentiation medium of ' Jiuzhouhong' was MS +0.8 mg/L 6-BA + 0.3 mg/L IAA + 3.0% sucrose + 1.3% agar strip, and adventitious buds could be induced after about 40 days with the bud formation rate of 61.2%. The speed of adventitious buds proliferation was the fastest in the MS + 1.0 mg/L 6-BA + 0.5 mg/L IAA + 3.0% sucrose + 1.3% agar strip medium with the proliferation coefficient of 3.62, and the buds grew strong and healthy. The best rooting medium of ' Jiuzhouhong' was I/2MS + 0. 6 mg/L IBA + 0.1 mg/L NAA + 3.0% su- crose + 1.3% agar strip, and the rooting rate was up to 91.2% with average 3.8 roots of each plant, and the growth of plant was strong, suit- able for transplanting. [ Conclusion] The study provided technical support for the large-scale production of the ' Jiuzhouhong' pomegranate.
