摘要
为了评价杜仲叶多酚体内和体外抗氧化活性,杜仲叶经大孔吸附树脂分离、有机溶剂萃取后,得到乙酸乙酯萃取物和正丁醇萃取物两种多酚纯化产物,测定各产物中多酚质量分数,并采用动物试验考察各纯化产物对小鼠丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和谷胱甘肽过氧化物酶(GSH-Px)活性的影响,以评价不同多酚产物的体内抗氧化活性,同时测定各产物清除羟自由基(.OH)、超氧阴离子自由基(O2-.)及1,1-二苯基-2-苦苯肼自由基(DPPH.)的作用,以评价不同多酚产物的体外抗氧化活性。结果表明:正丁醇萃取物中多酚质量分数高于乙酸乙酯萃取物;正丁醇萃取物具有更为显著的体内和体外抗氧化活性。
In order to study the antioxidation activity in vivo and in vitro of polyphenols separated from Eucommia ulmoides leaves,the Eucommia ulmoides leaves were separated by resin and organic solvent and then ethyl acetate extract and butyl alcohol extract were gotten.The polyphenols in the extracts were tested for content.The content of malondialchehyche(MDA),the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) were investigated in mice to evaluate the antioxidation activity in vivo.The scavenging capacity of the extracts on HO.,O2-.and DPPH.were tested to evaluate the antioxidation activity in vitro.The result showed that the content of polyphenols in butyl alcohol extract was higher than that in ethyl acetate extraction,and butyl alcohol extract showed more significant antioxidation activity in vivo and in vitro.
