摘要
目的:研究蟾酥水提取物对子宫颈癌HeLa细胞黏附、迁移和侵袭能力的抑制作用。方法:应用MTT法检测蟾酥水提取物对子宫颈癌HeLa细胞活力和细胞黏附能力的影响,划痕实验观察蟾酥水提取物对HeLa细胞运动能力的影响,Transwell小室模型研究蟾酥水提取物对HeLa细胞的迁移和侵袭的抑制作用,免疫印迹法检测基质金属蛋白酶-9(MMP-9)的表达。结果:100μmol/L或以上浓度的蟾酥水提取物作用24 h,Hella细胞生存率明显低于对照组(P均<0.05);20μmol/L或以上浓度的蟾酥水提取物作用24 h,HeLa细胞黏附率明显低于对照组(P均<0.05)。20μmol/L或以上浓度的蟾酥水提取物对Transwell小室膜上HeLa细胞的运动、迁移、侵袭均有抑制作用。50μmol/L蟾酥水提取物作用下,HeLa细胞的MMP-9蛋白表达强度比对照组明显减弱。结论:蟾酥水提取物能抑制HeLa细胞的黏附,阻止其迁移和侵袭,抑制MMP-9的活性可能是蟾酥水提取物体外诱导HeLa细胞发生凋亡的重要作用机制之一。
Objective : To investigate inhibitory effects of ~enenum Bufonis on the adhesion, invasion and mi- gration of human cervical carcinoma HeLa cell line. Methods: The viability of HeLa cells was measured by MTF assay. Cell adhesion assay, cell scratch assay and Transwell chamber assay were used to determine changes in ad- hesion, invasion and migration of HeLa cells after Venenum Bufonis treatment. Western blot was used to detect the expression of matrix metalloproteinase-9 (MMP-9). Results: The viabilities of HeLa cell treated 24h by 100 μmol/L or above Venenum Bufonis were lower than that of control group (all P 〈0. 05 ). And the adhesion rates of HeLa cell treated by 20 μmol/L or above Venenum Bufonis was lower than that of control group ( all P 〈 0. 05 ). 20 μmol/L or above Venenum Bufonis could obviously suppress the invasive and migrativeability of HeLa cells. Furthermore, MMP-9 protein expression of Hela treated by 50 μmol/L Venenum Bufonis was significantly de- creased than that of control group. Conclusion: It shows Venenum Bufonis can inhibit the adhesion, invasion and migration of HeLa cells, which may be associated with down-regu-lation of the expression of MMP-9.
