摘要
对苦丁冬青苦丁茶多糖(IKPS)的分离纯化及其单糖组成进行研究。IKPS粗品通过DEAE Sepharose FastFlow阴离子交换柱层析分离得到IKPS-1、IKPS-2、IKPS-3和IKPS-4共4个纯化组分,然后采用PMP柱前衍生化法(HPLC法)和糖腈乙酸酯衍生化法(GC法)分析IKPS及其纯化组分的单糖组成。结果表明:IKPS、IKPS-1、IKPS-2、IKPS-3和IKPS-4中均含有甘露糖、鼠李糖、半乳糖、阿拉伯糖、葡萄糖醛酸和果糖,除组分IKPS-4外的其他组分均含有葡萄糖;另外,IKPS和IKPS-1中还含有岩藻糖,IKPS、IKPS-1和IKPS-4检测到了半乳糖醛酸。2种方法检测结果基本一致,但HPLC法能检测到多糖组成中的果糖和糖醛酸信息,结果更为全面、可靠。
In the present study, crude polysaccharides from Ilex kudincha C.J. Tseng (IKPS) leaves were prepared and purified by DEAE Sepharose Fast Flow anion-exchange column chromatography. As a result, four purified fractions were obtained and named as IKPS-1, IKPS-2, IKPS-3 and IKPS-4. The monosaccharide composition of the fractions was analyzed by two different methods: HPLC with pre-column derivatization with 1-phenyl-3-methyl- 5-pyrazolonde (PMP) and GC after derivatization to aldononitrile acetates. All these purified polysaccharides were mainly composed of mannose, rhamnose, galactose, arabinose, glucuronic acid and fructose. Except IKPS-4, all other fractions contained glucose. Furthermore, IKPS and IKPS-1 also contained fructose, and IKPS, IKPS-1 and IKPS-4 contained galacturonic acid. The results of HPLC analysis were generally consistent with those of GC analysis. However, compared to GC analysis, HPLC analysis could provide more comprehensive and reliable information about the monosaccharide composition of polysaccharides from Ilex kudincha C.J. Tseng leaves.
