摘要
目的:研究玉郎伞多糖(YLSP)对Aβ25-35诱导PC12细胞凋亡的保护作用。方法:采用Aβ25-35诱导PC12细胞损伤凋亡模型,给予YLSP预处理后,MTT比色法分析细胞存活率,倒置相差显微镜、透射电镜观察细胞形态改变,以观察药物的保护作用。结果:MTT法显示与正常对照组相比较,模型组细胞存活率显著下降(P<0.01),与模型组比较,YLSP各浓度组均使细胞存活率显著升高(P<0.05或P<0.01);倒置相差显微镜、透射电镜观察显示Aβ25-35可导致PC12细胞凋亡,经YLSP处理后,模型细胞凋亡情况明显改善。结论:YLSP对Aβ25-35诱导的PC12细胞凋亡有保护作用。
OBJECTIVE To investigate the protective effect of YLS polysaccharides(YI.SP) on Aβ25-35-induced apoptosis in PC12 cells. METHODS PC12 ceils were treated with Aβ25-35 to establish a cell model of Alzheimer's disease. After pretreated with YLSP,MTT assay was employed to analyze the PC12 cell viability, the cellular morphology was observed with inverted phase contrast microscope and electron microscopy. RESULTS Aβ25-35 induced a decrease in viability of PC12 cells (P〈0. 01), When PC12 cells were co treated with YLSP and Aβ25-35, a decrease in viability of PC12 cells induced by Aβ25-35 was blocked by YLSP(P〈0. 01 or P〈0. 05). Morphological evidences for increasing apoptosis were observed in PC12 cells incubated in the presence of Aβ25-35, but the ceils treated with YLSP revealed decreasing apoptotic rates,it demonstrated that YLSP had a signifi- cant protective action against Aβ25-35 evoked apoptosis. CONCLUSION YLSP can resist the adverse effects of Aβ25-35 on increas- ing apoptotic rates,and it has the protective function towards nerve cells.
