间质性膀胱炎中膀胱上皮共表达干细胞因子和白细胞介素-6及其意义 被引量:10

Co-expression of stem cell factor and interleukin 6 in interstitial cystitis/bladder pain syndrome and their relationship with mast cells

摘要 目的检测问质性膀胱炎/膀胱疼痛综合征(IC/BPS)中干细胞因子(SCF)、白细胞介素-6(IL-6)和肥大细胞特异性类胰蛋白酶(tryptase)在膀胱上皮中的表达及定位,探讨其相关性及其与肥大细胞增殖的关系。方法采用免疫组织化学法检测SCF、IL-6及tryptase在IC/BPS和正常膀胱上皮组织中的表达及定位;采用实时定量聚合酶链反应(Real—timePCR)检测IC/BPS和正常膀胱上皮组织中上述3种基因mRNA的表达差异;Westernblot检测SCF、tryptase蛋白水平的表达差异。结果在IC患者膀胱上皮组织中SCF、IL-6和tryptase在mRNA及蛋白水平的表达均显著高于对照组(P〈0.05);在IC患者膀胱上皮组织中肥大细胞明显增多,并且表现出与膀胱黏膜层的密切关系,此外在mRNA水平SCF表达量与txyptase表达量呈正相关(r=0.844,P〈0.01)。结论Ic患者膀胱上皮组织中肥大细胞的增多和激活,可能是由于膀胱黏膜层共表达SCF和IL-6升高过度刺激的结果。 Objective To investigate the expression of stem cell factor (SCF), interleukin 6 ( IL-6 ) and tryptase in bladder epithelia, and their relationship with mast cells in interstitial cystitis/Had- der pain syndrome (IC/BPS). Methods The protein expression levels and localizations of SCF, IL-6 and tryptase in [C/BPS and normal bladder epithelial tissues were determined by using immunohistochemistry. The mRNA and protein expression levels of SCF, IL-6 and tryptase were detected by using real-time quanti- tative polymerase chain reaction (PCR) and Western blotting in IC/BPS and normal bladder epithelial tis- sues respectively. Results The mRNA and protein expression levels of SCF, IL-6 and tiTptase in IC/BPS bladder epithelial tissues were higher than those in normal bladder epithelial tissues ( both P 〈 0. 05 ). The number of mast cells in IC/BPS bladder epithelia tissues was significantly greater than in normal bladder epithelial tissues, and mast cells were found in close proximity of bladder epithelial cells. In addition, the mRNA expression levels of SCF and tryptase displayed a strong positive correlation (r = 0. 844 ,P 〈 0. 01 ). Conclusion The responses of mast cells in IC/BPS may be the result of hyperstimulation by local bladder epithelial SCF and IL-6 co-expression.
