siRNA沉默RAD18抑制结肠癌SW480细胞FANCD2蛋白泛素化并增强其对丝裂霉素C的敏感性 被引量:4

摘要 目的:研究RAD18特异性siRNA对结肠癌SW480细胞FANCD2泛素化和对化疗药物丝裂霉素C敏感性的影响。方法 :RAD18特异性siRNA转染SW480细胞后用丝裂霉素C处理,通过Western blot检测细胞RAD18及FANCD2泛素化的表达,MTT法检测干扰RAD18后细胞对丝裂霉素C的敏感性。结果:RAD18特异性siRNA转染SW480细胞96 h后,RAD18基因的表达明显降低,导致经丝裂霉素C处理后FANCD2泛素化表达明显降低;siRAD18组细胞对丝裂霉素C的敏感性显著增强。结论 :抑制RAD18蛋白表达能延缓结肠癌细胞FANCD2泛素化表达,并增强SW480细胞对丝裂霉素C的敏感性。
