摘要
目的观察法舒地尔对大鼠弥漫性轴索损伤后Rho激酶表达的影响,探讨其可能的机制。方法将60只大鼠随机分为3组:A组(假手术组,12只)、B组(模型组,24只)和C组(法舒地尔治疗组,24只),术后1 h、12 h、24 h及72 h 4个时间点处死大鼠,取脑组织进行Rho激酶表达的检测,应用末端转移酶标记技术(TUNEL)法观察神经细胞凋亡率。结果 B组、C组各时间点Rho激酶表达均高于A组(P<0.05),术后72 h最为明显,C组Rho激酶各时间点表达较B组减少(P<0.05)。结论 Rho激酶的表达与弥漫性轴索损伤密切相关,法舒地尔可有效抑制弥漫性轴索损伤后Rho激酶的激活,降低神经细胞凋亡率,减轻神经功能损伤。
Objective To study the effects of Fasudil on Rho kinase expression in diffuse axonal injury rats,and explore the possible mechanisms.Methods A total of 72 adult healthy male Spragne-Dawley(SD) rats were equally divided into sham operation group(group A),diffuse axonal injury group(group B),Fasudil treatment group(group C).The rats were sacrificed at 1 h,12 h,24 h and 72 h after injury,respectively.The brain tissues were removed to paraffin section.The expressions of Rho kinase were observed by the method of immunohistochemical technique.TUNEL method was used to observe the rate of neuronal apoptosis.Results The expressions of Rho kinase in group B and C,which increased significantly at 72 h,were higher than group A(P 0.05).The expression in group C was lower than that of group B at each time point(P 0.05).The expression in group C was lower than group B(P 0.05).Conclusion There was some difference in Rho kinase expression between the normal group and the diffuse axonal injury group.Fasudil could inhibit the activation of Rho kinase effectively and decreasing the cell apoptosis after diffuse axonal injury rats so as to achieve to protective effect of neuron.
