摘要
目的探讨富血小板血浆(platelet-rich plasma,PRP)对体外培养的根尖乳头干细胞(stem cells fromapical papilla,SCAP)生物学效应的影响。方法将SCAP细胞分别在标准培养液、PRP培养液、矿化诱导液及PRP+矿化诱导液中培养。通过缩胆囊素八肽(cell counting kit-8,CCK-8)法、碱性磷酸酶活性(alkaline phosphatase activity,ALP)检测、Western Blotting检测SCAP增殖及分化能力的改变。结果 PRP组及PRP+矿化诱导液组SCAP的增殖能力强于其他组(P<0.05)。与其他组相比较,PRP+矿化诱导液组SCAP的ALP活性及牙本质涎蛋白(dentin sialophosprotein,DSP)表达水平明显上调,矿化能力显著提高(P<0.05)。结论 PRP可明显促进SCAP细胞的增殖能力,与矿化诱导液联合应用对SCAP的矿化能力具有促进作用。
Objective To observe the effects of platelet-rich plasma on the biology effects of stem cells from apical papilla in vitro. Methods SCAP were treated with a standard PRP medium , mineralization-inducing medium and PRP + mineralization-inducing medium . Proliferation of SCAP was evaluated by CCK-8 eolorimetric assay. Alkaline phosphotase (ALP) activity was determined by enzyme kinetics methods. Western blotting were performed to investigate the mineralization ability of SCAP. Results The proliferation activity of PRP and PRP+ mineralization- inducing medium treated SCAP were significantly higher than other groups (P〈0.05). ALP activity and DSP levels of the PRP+ mineralization-inducing medium group was significantly higher than other groups (P 〈0.05). Conclusion PRP significantly promotes the proliferation of SCAP, and PRP+ mineralization- inducing medium significantly promotes the mineralization of SCAP.
