慢性阻塞性肺疾病患者外周血单个核细胞糖皮质激素抵抗的体外观察 被引量:6

摘要 慢性阻塞性肺疾病(chronic obstructive pulmonarydiease,COPD)是以气道、肺部及全身炎症为特征的疾病,吸烟及有害颗粒的吸入是导致炎症发生发展的主要原因。糖皮质激素是目前常用的治疗COPD的抗炎药物,但临床疗效不满意,可能与COPD组蛋白去乙酰化酶(histone deacetylases,HDACs)活性下降导致的糖皮质激素抵抗有关[1-2]。既往的研究表明红霉素可以增加氧化应激下单个核细胞HDAC的活性,并减少NF-κB转录活性及IL-8的合成[3]。因此,本实验采用分离的外周血单个核细胞(peripheralblood mononuclear cell,PBMC),建立糖皮质激素抵抗的体外细胞模型,从而为进一步寻找有效的减轻COPD糖皮质激素抵抗的药物及其机制研究提供基础。
