咬合力与大鼠牙周组织改建关系研究中动物模型的建立 被引量:18

摘要 目的 :研究在体内情况下咬合力影响牙周组织改建的分子机制。方法 :采用拔牙法拔除大鼠一侧上颌磨牙 ,建立模拟下颌磨牙咬合力丧失及增强的动物模型。选择某一特定细胞因子 ( IL- 6 )用于免疫组化、原位杂交实验中鉴定 ,且行组织形态学观察 ( HE染色 )。结果 :免疫组化以及原位杂交结果显示 ,IL- 6蛋白及 IL- 6 m RNA表达在咬合力增强组及减弱组均较正常咬合力组明显改变。组织形态学观察表明 ,咬合力增强及减弱对牙周组织改建的影响与正常咬合力明显不同。结论 :通过拔除大鼠一侧上颌磨牙建立的动物模型 ,达到较好地模拟咬合力丧失、增强的目的。此动物模型有效。 ?Objective: To establish an animal model for the study of molecular mechanism of the effects of bite force on periodontium remodeling. Methods: Left maxillary molars of rats were extracted and the left mandibular molar area was used as the bite force loss; right mandibular molars served as bite force increase. Periodontium remodeling on both sides was observed with HE staining, immunohistochemistry and in situ hybridization techniques 1,2,3 and 4 weeks after the extraction. Results: Decrease of periodontal space was observed in the group of bite force loss and increase in the group of bite force increase at all the intervals of observation (P<0.05).Increased IL6 and IL6 mRNA expression was found in the two tested groups. Conclusion: The animal model may be used to represent the bite force loss and bite force increase.
