解偶联蛋白2在反流性食管炎、Barrett食管和食管腺癌黏膜中表达差异的研究 被引量:7

摘要 目的胃食管反流病的发病率呈逐年增加的趋势,但其发病机制和疾病的演变过程仍不明确。文章探讨反流性食管炎(reflux esophagitis,RE)、Barrett食管(Barrett esophagus,BE)和食管腺癌(esophageal adenocarcinoma,EAC)中解偶联蛋白2(uncoupling protein 2,UCP2)表达水平及意义。方法分别用硫代巴比妥酸显色法和黄嘌吟氧化酶法测定对照组和各实验组患者血浆中丙二醛(malondialdehyde,MDA)和超氧化物歧化酶(superoxidedismutase,SOD)的含量。经胃镜取材,免疫组织化学方法分别检测各组患者食管黏膜组织中UCP2蛋白的表达。结果对照组、RE组、BE组和EAC组血浆MDA表达量呈增加趋势,各组差异有统计学意义(P<0.05),血浆SOD表达量逐渐降低,对照组和各实验组差异有统计学意义(P<0.05),但RE组和BE组以及BE组和EAC组差异没有统计学意义(P>0.05)。对照组无UCP2蛋白表达,RE组、BE组和EAC组UCP2蛋白的表达量逐渐增加,各组差异有统计学意义(P<0.05)。结论反流性食管炎和Barrett食管黏膜组织中UCP2蛋白的表达程度和食管腺癌的发生有一定的关系。 Objective The incidence of gastroesophageal reflux disease is on the rise, but its pathogenesis and evolution remain to be further defined. This article investigates the expression of uncoupling protein 2 (UCP2) in reflux esophagitis ( RE), Barrett esophagus (BE) and esophageal adenocarcinoma (EAC). Methods This study included 60 cases of RE, 45 cases of BE, 10 cases of EAC and 30 normal controls. We measured the levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in the plasma of all the subjects using the thiobarbituric acid cololimitric and xanthine oxidase methods. We also determined the expression levels of UCP2 in their esophageal mucosa tissues by immunohistochemistry. Results The level of plasma MDA was significantly increased in the RE, BE and EAC groups, each higher than the former (P 〈 O. 05 ), while that of SOD remarkably decreased, with significant difference between the controls and patients (P 〈 0.05), but not between RE and BE or BE and EAC groups (P 〉 0.05 ). The UCP2 expression was markedly elevated in the RE, BE and EAC groups, each higher than the former ( P 〈 0.05 ), but not observed in the normal control group. Conclusion The expression of UCP2 is correlated with the occurrence of esophageal adenocarcinoma.
