摘要
随着分子生物学技术的发展,HLA基因序列、氨基酸序列及蛋白质晶体空间结构逐步明确,使得可以运用计算机生物信息方法在移植术前HLA配型中发挥更为有效的作用。HLAMatchmaker软件是基于Excel软件开发,通过对供、受者HLA抗原氨基酸序列上的eplets进行比较,寻找合适的供者,提高移植术后受者生存率。运用此软件分析可避开基因错配位点,让群体反应性抗体高敏感性受者亦能接受移植。本文就HLAMatchmaker软件在HLA配型中的应用原理及前景展开综述。
With the development of molecular biology technology, the sequences of DNA and the amino acid and protein crystal structure of HLA are explicit. These advances facilitate the use of bioinformatics tools to play a more effective role in matching HLA between donors and recipients before transplantation. HLAMatchmaker is an Excel-based algorithm which determines donor-recipient HLA eplets of amino acid sequence, so as to find a suitable donor to improve the survival of allograft. In addition, HLAMatchmaker algorithm can avoid mismatch locus, and panel reaction antibody high sensitivity patients also can accept transplants. We reviewed the principles, and application prospect of HLAMatehmaker in transplantation.
