雷公藤内酯醇对糖尿病大鼠脑内葡萄糖转运蛋白表达的影响

摘要 目的探讨雷公藤内酯醇对糖尿病大鼠脑内葡萄糖转运蛋白(Glut-1、Glut-3)表达的影响。方法大鼠45只,分为正常对照组13只和糖尿病(DM)模型组32只。模型组每天一次腹腔注射链脲佐菌素(STZ)52 mg·kg-1,连续5 d;正常对照组仅腹腔注射等体积0.1 mol·L-1枸橼酸。将造模成功的28只大鼠随机分为模型对照组和雷公藤内酯醇组各14只。雷公藤内酯醇组灌胃给予雷公藤内酯醇混悬液1.8 g·kg-1,正常对照组和模型对照组则灌服等体积饮用水,每日上午1次,3组均连续给药8周。结果正常对照组Glut-1和Glut-3的表达海马部位多于皮质部位。模型对照组皮质部位及海马部位Glut-1、Glut-3变化不明显(P>0.05)。雷公藤内酯醇组皮质部位及海马部位Glut-1表达分别为(1.31±0.34)%,(1.03±0.25)%,均明显降低(P<0.01);海马部位Glut-3表达有所增加(P<0.01)。结论雷公藤内酯醇能明显降低糖尿病大鼠脑内的Glut-1含量。 Objective To explore the effects of triptolide on Glut-1 and Glut-3 expression in diabetic rat brain. Methods Forty-five rats were randomly divided into normal control group (n = 13 ) and diabetic model group (n = 32 ). In the model group, the rats were intraperitoneally injected with streptozotocin (STZ) 52 mg · kg-1 once daily for five consecutive days; in the normal control group, the rats were intraperitoneally injected with equal volume of citric acid 0.1 mol ·L-1. Twenty-eight DM rat models were successfully established and divided into model control group (n=14) and triptolide (TP) group (n = 14). Rats in the TP group were administered with triptolide suspension intragastrically at 1.8 g · kg -1 daily for 8 weeks, and those in the model control group were administered with equal volume of water. Results In normal control group, expression levels of Glut-1 and Glut-3 were higher in the hippocampus than in the cortex. In model control group, expression levels of Glut-I and Glut-3 in the hippocampus and cortex were not changed significantly ( P〉0.05 ). In TP group, Glut-1 expression in the cortex and hippocampus was ( 1.31 ± 0. 34 ) % and ( 1.03± 0. 25 ) %, significantly decreased ( P〈 0.01 ). Glut-3 in hippocampus was increased significantly (P〈0.01). Conclusion Triptolide can significantly lower the Glut-1 content in the diabetic rat brain.
