TNF-α对肠上皮细胞通透性的影响及其NF—κB/MLC—P调控途径的研究 被引量:3

摘要 【目的】探讨肿瘤坏死因子-α(TNF-α)对人肠上皮细胞问通透性的影响及作用机制。【方法】利用Caeo-2细胞建立肠上皮细胞屏障模型;分为对照组和NF—κB抑制组,分别为转染了空载质粒(vector)和DN—MuikBa质粒的Caco-2细胞,两组经TNF-α预处理后,应用电阻仪测定各组跨上皮细胞电阻(TEER)变化,罗丹明-鬼笔环肽直接染色观察细胞骨架改变,同时采用荧光素酶报告基因检测NF—κB活性,免疫印迹法检测肌球蛋白轻链(MLC)磷酸化水平。【结果】TNF-α活化NF-κB,引起F-actin(细胞骨架蛋白)重排,MLC磷酸化水平增高,细胞通透性增加;反之,抑制NF-KB活化,上述作用被明显抑制。【结论]TNF-α导致Caco-2细胞间的通透性增加,其机制可能与NF—κB活化,F—actin重排及MLC磷酸化有关。
