摘要
目的 :建立测定大鼠血浆样品中川芎哚浓度的气相色谱 -质谱 (GC -MS)定量分析方法。方法 :血浆样品经乙醚 -异丙醇 (10∶1)提取、硅烷化处理后 ,选择GC -MS选择性离子和辅助定量离子监测。结果 :本法线性范围为 2~ 6 0ng·mL-1,灵敏度为 1ng·mL-1,回收率达 82 % ,日内日间RSD均小于 9 0 %。结论 :本法为川芎哚临床药代动力学研究提供了准确、可靠的方法。
Objective:To develop a method for the determination of perlolyrine in rat plasma using GC-MS. Methods:Perlolyrine in plasma was extracted with ethyl ether—iso-propyl alcohol and determined by means of GC-MS with selected ion and qualifier ion monitoring.Results:The standard curve was linear between 2~60 ng·mL -1 and the minimum detection limit was 1 ng·mL -1 .The recovery of perlolyrine in plasma was above 82% and the relative standard deviation ( RSD ) less than 9 0%.Conclusion:It provided an important method for the determination of clinical pharmacokinetics.
