HPLC法测定泛昔洛韦含量

Determination of Famiciclovir by HPLC

目的 :用高效液相色谱法测定泛昔洛韦。方法 :在ODS柱上 ,以磷酸盐缓冲液 (0 0 2mol·L-1磷酸二氢钾 10 0 0mL ,加三乙胺 3mL ,用 85 %磷酸调节至 pH 2 5 ) -甲醇 (6 5∶35 )为流动相 ,于 2 2 0nm波长处检测 ,流速为 1 0mL·min-1;利巴韦林为内标 ,泛昔洛韦与内标的保留时间分别为 4 90min和 2 77min。结果 :浓度线性范围为 0 0 1～ 0 0 8mg·mL-1,重复性试验RSD =1 0 % (n =5 )。结论 :本法含量测定结果满意。

Objective:To establish the determination of the content of Famciclovir by HPLC.Method:The chromatographic condition were:Ultrashere C 18 5 μm(4 6 mm×250 mm);phosphate buffer(0 02 mol·L -1 potassium dihydrogen phosphate solution 1000 mL add triethylamin 3 mL,adjusted with 85% phosphoric acid to pH 2 5)—methanol(65∶35)as mobile phase;ribavirin as internal standard;flow rate was 1 0 mL·min -1 ,wavelength of the detector was 220 nm.Results:The retention time of Famciclovir and internal standard were 4 90 min and 2 77 min respectively The linear range of calibration curve was 0 01～0 08 mg·mL -1 ( r=1 000 0) .Conclusion:This method is suitable to determine Famciclovir
