摘要
目的 :建立兔血浆中钙拮抗剂尼卡地平的反相高效液相色谱分析方法 ,并对其在兔体内的药代动力学进行研究。方法 :血浆样品在弱碱性条件下经环己烷 -异丙醇 (96∶4)提取 ,用LiChrosorbC18反相柱 ,甲醇 -水(75∶2 5 )为流动相 ,流速 1 0mL·min-1,检测波长 2 36nm等条件下 ,分析测定。结果 :方法平均回收率为98 2 7%~ 10 0 4% ,RSD <5 0 % ,线性范围 :11~ 110ng (r =0 9981) ,最低检测限为 10ng。 结论 :本法操作简便、准确灵敏、重现性好 ,可用于尼卡地平的药动学研究。
Objective:To establish HPLC method for determination and pharmacokinetic study of nicardipine in rabbit.Methods:The blood samples were diluted by 0 05 mol·L -1 Na 2HPO 4 buffer solution(pH=7 4) and extracted with cyclohexane-2 propanol(96∶4).The LiChrosorb C 18 column(150 mm×4 6 mm,5 μm)was used as analytical column.The mobile phase consisted of methanol-water(75∶25).The flow rate was 1 0 mL·min -1 with UV detection at 236 nm.Results:The recovery of the method was 98 27%~100 4%, RSD <5 0%.The linear range was 11~110 ng( r =0 998 1) and determination limit was 10 ng.Conclusion:The method had a good selectivity、sensitivity and reproducibility and can be used for pharmacokinetic studies of nicardipine in rabbit plasma.
