大鼠大脑中动脉近端及远端阻塞脑缺血模型的脑梗死体积和行为学差异比较 被引量:5

The Effect of Proximal and Distal Middle Cerebral Artery Occlusion on Cerebral Infarction Volume and Behavioral Outcome in Rat

摘要 目的建立两种稳定的大鼠大脑中动脉近端和远端闭塞的脑缺血模型,比较两种方法在脑梗死体积、行为学等方面的差异。方法 58只大鼠随机分为线栓组(传统的MCAO模型制作,n=24),远端大脑中动脉闭塞组(dMCAO,电凝一侧大脑中动脉远端和结扎双侧颈总动脉,n=24),对照组(未予任何手术处理,n=10)。模型制作后进行行为学测验并计算脑梗死体积。结果脑梗死体积比较线栓组与dMCAO组,差异有显著统计学意义(P<0.01);dMCAO组的行为学影响较小。结论相对于线栓法大鼠模型,dMCAO模型易制作、梗死体积较小、以皮质区损伤为主,对运动功能的影响较线栓法脑缺血模型小。 Aim To compare the differences of infarct volume and neurological behavior outcome between proximal middle cerebral artery occlusion (suture MCAO) model and distal middle cerebral artery occlusion (dMCAO) model. Methods Rats were randomly divided into 3 groups: suture MCAO (n=24), dMCAO (n=24) and sham-operated group (n= 10). Infarction volume was measured and behavior tests were performed at different durations after cerebral ischemia. Results Significant differences were found in the infarction volume between the suture MCAO and dMCAO group (P〈0.01). In addition, based on the beam walking and elevated body swing tests, significant differences were also found between the suture MCAO and the control group, between dMCAO group and the control group(P〈0.01), but no significant differences were found between MCAO and dMCAO (P〉0.05). There were significant differences between the MCAO and dMCAO group (P〈0.01) in the cylinder test. Conclusion Compared to MCAO model, dMCAO model was easily performed, and its infarct volume, mainly in cortex, was smaller. The neurological behavior deficiencies were less severe.
