镉对大鼠睾丸生精细胞凋亡的影响及锌的保护作用 被引量:8

摘要 为探讨镉对睾丸结构和功能损害及锌对其保护作用的分子机制 ,研究镉对生精细胞凋亡的影响和锌的保护作用 ,选用 2月龄 Wistar雄性大鼠 2 4只 ,均分为染镉组、镉加锌组和对照组 ,分别自腹腔注射氯化镉 (2 mg/ kg BW) ,注射氯化镉同时及其前后 2 h各注射醋酸锌一次和等量生理盐水 ,7d后采用原位缺口平移技术 (INST)检测各组睾丸生精细胞凋亡数目的变化。结果显示与对照组相比 ,染镉组大鼠睾丸生精细胞凋亡数目明显增多 (P<0 .0 1,t=3.87) ;镉加锌组睾丸生精细胞凋亡数目无明显变化 (P>0 .0 5 ,t=1.34 )。染镉后大鼠睾丸生精细胞凋亡明显增加 ,锌对镉所诱导的生精细胞凋亡有保护作用 ,这些变化可能是镉损害睾丸结构和功能以及锌对其起保护作用的分子机制之一。 To study the effect of cadmium on the apoptosis of spermatogenic cells of rat testis and the protective effect of zinc against it. 24 Wistar male rats were divided into 3 groups (cadmium, cadmium and zinc, and contral rats). Rats were injected with low dose CdCl 2(2mg/kgBW),zinc acetate(ZnAc,15mg/kgBW) before and after injected with low dose CdCl 2 and ZnAc(50mg/kgBW). The contral rats were injected with same dose of 0 9%NaCl.After 7 days, the apoptosis of spermatogenic cells was studied with in situ nick translation(ISNT) technique. Compared with contral group, the number of apoptosis spermategenic cells was obviously increased in cadmium group( P <0 01, t =3 87), but it was not significantly different in cadmium and zinc rats. Cadmium treatment could accelerate testis apoptosis. Zinc could provent it.
