摘要
目的 观察恒定磁场诱导下阿霉素磁性蛋白微球体外对恶性肿瘤细胞生长的抑制作用。方法 体外培养的Walker 2 5 6细胞分为阿霉素磁性微球联合磁场组 ,阿霉素磁性微球组及阿霉素组三组 ,倒置显微镜观察细胞的生长状态 ,改良MTT法检测各组细胞的抑制率及半数抑制剂量 ,3H TdR掺入法检测各组抑制率。结果 阿霉素磁性微球组与游离阿霉素组抑制率及半数抑制剂量相似 (P >0 .0 5 ) ,联合磁场组抑制率明显提高 ,半数抑制剂量约降低一半 (P <0 .0 1) ,两种检测方法的药敏试验相符率达 84.19% ,细胞形态出现显著变化。结论 阿霉素磁性蛋白微球与游离阿霉素对恶性肿瘤细胞具有相似的抑制作用 ,联合恒定磁场可增强其抑瘤效应。
Objective To study the inhibitory effects of the adriamycin magnetic albumin microspheres(ADM-MAMs) to the Walker-256 malignant tumor cells in vitro induced by the constant magnetic fields.Methods The cultured Walker-256 cells were divided into three groups:group of ADM-MAMs combined with magnetic fields,groups of ADM-MAMs without magnetic fields and ADM group.The growth states of the cells were observed and photographed under the inverted microscope.The inhibitory rate(IR) and 50% inhibitory concentration(IC 50 ) were assayed by the modified MTT colorimetric method and 3H-TdR involved method.Results The IR and IC 50 of the ADM-MAMs group were similar to those of the ADM group(P> 0.05 ),but the group of ADM-MAMs combined with the magnetic fields had the obviously increased IR and IC 50 as well as the magnificent changes of the cells' growing shapes.Conclusion The anticancer effects of ADM-MAMs to the maligant tumor cells in vitro were similar to that of ADM and could be increased by combination with the permanent magnetic fields.
