摘要
建立高效液相色谱-内标法测定奥拉西坦的含量。以咖啡因为内标物,Luna NH2(4.6 mm×250 mm,5μm)为分析柱,乙腈∶水(95∶5)为流动相,检测波长220 nm,柱温30℃。结果表明该方法精密度和回收率良好,奥拉西坦质量浓度浓度在0.04~0.8(mg/mL)范围与其峰面积内呈良好线性关系,r=0.999 1。该法与凯氏定氮法测定结果一致,操作简便、快速、可靠,便于奥拉西坦的质量控制。
A High Performance Liquid Chromatography (HPLC) method with caffeine as an internal standard was developed for determination of oxiracetam. The sample was determined by using Luna NH2 (4. 6 mm ×250 mm,5μm) column, acetonitrile: water (95: 5) as a mobile phase. The column temperature was at 30 ℃. UV de- tection wavelength was set at 220 nm. Results indicated that the method had good accuracy and recovery rate. The standard curve was linear over the range of 0.04 - 0. 8 mg/mL, r = 0. 999 1. It is conclude that the HPLC de- termination of oxiracetam is consistent with the Kjeldah' s method, and also it is convenient, rapid, reliable.
