高效液相色谱法测定不同产地大腹皮中槟榔碱的含量

HPLC Determination of Arecoline in Arecae Pericarpium from Different Habitats

目的建立高效液相色谱法测定大腹皮中槟榔碱的含量。方法采用Thermo-BioBasic SCX离子交换柱(4.6 mm×250mm,5μm),流动相为乙腈-0.2%磷酸溶液(55∶45),浓氨试液调节pH值至3.8,流速1.0 mL.min-1,柱温30℃,检测波长215 nm。结果氢溴酸槟榔碱的色谱峰面积与质量浓度呈良好的线性关系,线性范围为5.2～83.2μg.mL-1(r=0.999 1),平均回收率(n=9)为98.8%。结论该方法简便、准确、专属性强,为大腹皮饮片的质量评价提供了方法。

OBJECTIVE To establish a quantitative determination method for arecoline in Arecae Pericarpium. METHODS The determination was performed on thermo-biobasic SCX column （4. 6 mm ×250mm,5μm） with mobile phase consisting of acetonitrile-phosphoric acid solution （55：45）. The flow rate was 1.0 mL ·min-1, the column temperature was 30 ℃, and the detection wavelength was 215 nm. RESULTS The method had good linear relationship for areeoline hydrobromide in the range of 5.2 - 83.2 μg·mL-1 （ r = 0. 999 1 ）. The average recovery （ n = 9 ） was 98.8% for arecoline hydrobromide. CONCLUSION The method is simple, accurate and specific. It provides a reliable way for evaluating the quality of Arecae Pericarpium.
