摘要
目的采用HPLC法测定假蒟中α-细辛脑的含量,观察其动态积累变化。方法色谱柱为phenomenex ODS2柱(250mm×4.60 mm,5μm),流动相为乙腈-0.05%磷酸溶液(49∶51),流速1.0 mL.min-1,检测波长313 nm,柱温30℃。结果α-细辛脑0.0975~1.9500μg与峰面积的线性关系良好(r=0.9998),平均回收率为101.59%(RSD=2.10%,n=6)。结论所用方法简便、快捷、重复性好,可作为假蒟药材中α-细辛脑的含量测定方法。
OBJECTIVE To determine α-asarone in Piper sarmentosum Roxb.by HPLC,and observe the changes of dynamic accumulation of α-asarone.METHODS The chromatographic column was C18 ODS2(250 mm×4.60 mm,5 μm),and the mobile phase was acetonitrile-0.05% H3PO4(49:51)with a flow rate of 1.0 mL·min^-1,the column temperature was 30 ℃ and the detection wavelength was at 313 nm.RESULTS There was a good linear relationship for α-asarone during 0.0975-1.9500 μg(r=0.9998).The average recovery rate was 101.59%(RSD=2.10%,n=6).The dynamic accumulation of the content of α-asarone was higher in autumn and winter.CONCLUSION The method is simple,fast and reproducible,and can be used for determination of α-asarone in Piper sarmentosum.
