芜菁子中β-谷甾醇的TLC鉴别及含量测定 被引量:2

摘要 目的定性鉴别芜菁子中的β-谷甾醇及含量测定。方法采用TLC法及HPLC法。以石油醚-乙酸乙酯(7∶3)为展开剂在硅胶GF254板上展开。色谱柱为YMC-PackODS-A(250 mm×4.6 mm,5μm),流动相为甲醇-乙腈-水(55∶9∶36),检测波长210 nm,流速1.0 mL.min-1。结果β-谷甾醇0.22~8.8μg与峰面积线性关系良好(r=0.9990),平均回收率为99.2%,RSD=1.6%。结论所用方法简便准确,重复性好,专属性强,可用于芜菁子的质量控制。 OBJECTIVE To establish the quality standards of the seeds of Brassica rapa L..METHODS TLC and HPLC was used to qualitatively identify and determine the content of β-sitsterol of 10 block of the seeds.Petroleum ether-ethyl acetate(7:3)was used as a developing solvent on Silica gel GF254 plates.The separation was performed on C18 column with methanol-acetonitrile-water(55:9:36) as a mobile phase at a flow rate of 1.0 mL·min^-1.The detection wavelength was 210 nm at 25 ℃.RESULTS There was a good linear relationship between the mass and the peak areas of β-sitsterol in the ranges of 0.22-8.8 μg(r=0.9990).The average recovery was 99.2% with RSD of 1.6%.CONCLUSION The method is accurate and reproducible,and can be used to control the quality of the seeds of Brassica rapa L..
