如意珍宝片对脑缺血大鼠后遗症的改善作用 被引量:4

摘要 目的:研究如意珍宝片对永久性大脑中动脉栓塞致大鼠局灶性脑缺血(pMCAO)的影响。方法:以线栓永久性阻断大脑中动脉致大鼠局灶性脑缺血,(1)SD大鼠动物随机分两组,模型组和如意珍宝片1 g.kg-1,术前给药3 d,术后连续给药,分别于术后8 d、14 d、21 d、28 d、42 d进行加速转棍实验,术后20 d进行平衡木实验,术后24 d进行步态实验,术后42 d以TTC染色法检测脑梗死体积;(2)SD大鼠动物随机分4组,假手术组、模型组、如意珍宝片0.5 g.kg-1组、如意珍宝片1 g.kg-1组,缺血24 h进行动物神经行为评分及脑组织匀浆中TNFα的检测。结果:与模型组相比,缺血24 h后如意珍宝片1 g.kg-1组动物神经行为评分下降17%,脑组织匀浆中TNFα含量减少55%,且42 d加速转棍持续时间延长159%(P<0.05),平衡木持续时间延长71%(P<0.01),后肢不对称性显著改善(P<0.05),脑梗死体积减少41%(P<0.01)。结论:如意珍宝片对脑缺血后的运动障碍具有一定的改善作用。 Objective: To investigate the protective effect of Ruyi Zhenbao Pian in focal cerebral ischemic rats. Methods: Focal cerebral ischemia in male SD rats was induced by permanent middle cerebral artery occlusion (pMCAO). 24 h after ischemia in rats, neurological deficit scores were evaluated, and the content of TNF a in ischemic area was detected. The behavioral ability was evaluated by the accelerated rotarod test, balance beam test and footprint test. The TTC staining was used to evaluate the infarct volume after 42 d of MCAO. Results : After 24 h of MCAO, the TNF a and neurological deficit scores were reduced in Ruyi Zhenbao Pian group. Compared with model group, Ruyi Zhenbao Pian at dose of 1 g . kg-1 increased remarkably the duration on the rotarod and the balance beam, decreased significantly the infarct volume. Conclusion: Ruyi Zhenbao Pian has potential neuroprotective action against the sequela after the focal cerebral ischemia in rats.
