摘要
目的:探讨心肌梗死后1 d与7 d穴位移植MSCs对梗死区促血管新生因子及血管新生的影响。方法:用开胸结扎冠状动脉前降支的方法建立急性心肌梗死模型,将已建立心肌梗死模型的日本大耳白兔随机分为正常组、模型对照组、穴位移植干细胞1 d组、穴位移植干细胞7 d组。抽取健康日本大耳白兔骨髓,分离培养骨髓间充质干细胞并传代,穴位注射干细胞组于急性心肌梗死形成后1 d与7 d,取心俞(双侧)、至阳、膻中4穴,穴位注射干细胞混悬液。免疫组化法检测梗死心肌组织CD31、VEGF平均光密度值。结果:CD31平均光密度与正常组相比较,穴位移植干细胞1 d与7 d组均降低,P<0.01;与模型组比较,穴位移植干细胞1 d组表达上调但无统计学差异,穴位移植干细胞7 d组上调P<0.01;穴位移植干细胞1 d组与穴位移植干细胞7 d组比较,穴位移植干细胞7 d组CD31表达上调P<0.01。BFGF平均光密度值,与正常组相比较,穴位移植干细胞1 d组表达下调,P<0.01,穴位移植干细胞7 d组表达上调,P<0.05;与模型对照组比较,穴位移植干细胞1 d组表达下调,P<0.01,穴位移植干细胞7 d组上调但无统计学差异;穴位移植干细胞1 d组与穴位移植干细胞7 d组比较,穴位移植干细胞7 d组BFGF表达上调P<0.01。结论:穴位注射骨髓间充质干细胞7 d组可使梗死区及周围组织CD31表达上调,诱导梗死区及周围组织血管新生,其机制是刺激心肌组织中bFGF表达上调。
Objective : T9 explore one day and seven days after myocardial infarction with MSCs points transplantation to promote angiogenic factors and the effect of angiogenesis. Methods : With open thoracio ligation anterior descending coronary artery method, the model of acute myocardial infarction was established. The constructed model rabbits of myocardial infarction were randomly divided into 3 groups,including model control group, point injecting stem cells for 1 day group, point injecting stem ceils for 7 days. Bone marrow has been isolated, cultured, and passed down. The points of Xinshu (bilateral) ,Zhiyang and Danzhong were injected stem cells 1 and 7 days after AMI. The immunohistochemical method was used to detect average light density values of CD31 and VEGF. Result: Compared with normal group CD31 average light density in point injection 7 days and 1 day groups were both lower( P 〈 0. 01 ) ;compared with model group, that in point injection 1 day group increased without statistical significance,in point injection 7 days group raised (P 〈 0.01 ) ; compared with point injection 1 day group, CD31 expression in point injection 7 days group increased(P 〈 0. 01 ). Compared with the normal group, BFGF average light density value in point injection 1 day group decreased (P 〈 0. 01 ), in point injection 7 days group raised (P 〈 0.05 ) ; compared with the model group, that in point injection 1 day group decreased and in 7 days group increased without significance(P 〈 0.01 ). 1 day group compared with 7 days group, BFGF in 7 days group increased(P 〈0. 01 ). Conclusion:Point injection of bone marrow mesenchymal stem cells 7 days group can make CD31 expression increase in the infarction area and surrounding tissues and induce angiogenesis, and its mechanism is to stimulate the myocardial tissue to increase bFGF expression.
