2型糖尿病大鼠造模比较及其脂质过氧化损伤探究

Exploration of Damage due to Lipid Oxidation and Comparison of Establishing Type 2 Diabetes Rat Model

目的:通过STZ联合高糖高脂饮食喂养诱导出稳定的2型糖尿病大鼠模型。方法:将90只雄性SD大鼠,随机分成空白组(18只)和造模组(72只),并按照不同STZ注射方式及高脂高糖饮食喂养先后分成4个实验组,最终检测FBS、TC、TG、HDL、LDL等指标进行考察,以血糖值FBG稳定在7.0～18 mmol/L为造模成功。结果:高脂高糖喂养后一次性STZ尾静脉注射成模率较满意。结论:通过本试验方法可以建立出比较稳定的2型糖尿病大鼠模型,符合人2型糖尿病特点,为进一步实验研究2型糖尿病奠定基础。

Objective：To induce a stable rat model of type 2 diabetes via STZ injection after feeding with high sugar and fat diet. Methods ：90 male Sprague Damley （SD）rats were randomly divided into normal control group （NC, n = 18 ） and model group（MG,n =72）. MG were injected with STZ in different methods and high sugar and fat diet order,then fasting blood - glucose （ FBG）, triglycerides （ TG）, total cholesterol （ TC ）, high density lipoprotein （ HDL）, low density lipoprotein（LDL） and other indexes were tested. The successful rat model was determined by the standards that FBG range was 7.0 -18 mmol/L. Results ：Intravenous injection with STZ after high sugar and fat diet can establish type 2 diabetes rat model with high successful rate. Conclusion：We can induce a T2DM rat model with this method, contributing to further experimental research of T2DM.
