大黄游离蒽醌的大孔吸附树脂纯化工艺研究 被引量:5

摘要 目的:筛选大黄游离蒽醌的大孔吸附树脂纯化最佳工艺。方法:通过静态吸附和动态洗脱实验,以吸附率为指标,考察6种大孔树脂对大黄游离蒽醌的吸附性能,评价不同大孔树脂对大黄游离蒽醌的富集纯化能力,确定最佳型号的树脂及洗脱参数。结果:6种树脂中,AB-8型树脂对大黄游离蒽醌的吸附能力和解析能力最强,实验确定的最佳工艺条件为:含生药量为0.08 g/mL的大黄提取溶液1 BV.h-1流速上样,以水、30%、50%、70%、90%乙醇各4BV洗脱,收集50%、70%、90%乙醇的洗脱液,合并,所得大黄游离蒽醌含量达81.91%。结论:AB-8型树脂对大黄游离蒽醌具有良好的富集纯化作用。 Objective:To investigate optimal purification technology of free anthraquinones in Rhei Radix et Rhizoma on chromatograph of macroporous resin. Methods:With :adsorption rate and desorption rate as indexes, purifaction capacities of six types of macroporous resins were evaluated to free anthraquinones in Rhei Radix et Rhizoma by static state adsorption and dynamic state elution experiments, to determine the elution parameter and the optimal macroporous resins. Result:AB- 8 macroporous resins showed well separated and purified actions for free anthraquinones in Rhei Radix et Rhizoma. The optimal purification technology conditions were determined as follows :the solution with 0.08 g/mL dose of erud drug was subjected to the column of AB -8 macroporous resins at 1BV . h-1 of flow rate, eluted with 4BV of water, 30% ,50% ,70% and 90% ethanol solution,then 50% ,70% and 90% ethanol elution were collected and concentrated. The content of free anthraquinones in Radix et Rhizoma Rhei was 81.91%. Conclusion:AB -8 macroporous resins show a satisfactory purification to free anthraquinones in Rhei Radix et Rhizoma.
