肝癌干细胞标志物CD44在化学诱癌过程中的变化及意义 被引量:4

摘要 目的检测CD44在化学诱导C57BL/6J小鼠肝癌过程中各时期的动态变化。方法以45只正常C57BL/6J雄性小鼠为对照组,通过化学法诱发50只C57BL/6J雄性小鼠肝癌(诱癌组)。观察小鼠成瘤情况和生长状态,对每2周定期处死小鼠获得的组织标本分别进行病理学切片观察,实时定量PCR(real-time PCR)技术检测小鼠肝癌组织及相应阶段正常组织CD44 mRNA的表达水平,蛋白质印迹法、FCM法和免疫组化法检测CD44的蛋白表达情况,应用FCM法定量检测细胞表面抗原的表达情况。结果化学诱导20周后,成功诱发小鼠肝癌。病理学检测显示:诱癌组小鼠早期主要表现为肝组织充血水肿,伴有少量变性坏死,为药物性肝炎的表现,同时部分区域可见纤维组织增生;16周后肝细胞组织表面开始出现灰白色结节,数目不等,镜下可见肝细胞非典型增生;20周时肿瘤结节明显增大,最大直径可达25 mm,镜下显示为高分化肝细胞癌。real-time PCR、蛋白质印迹及FCM检测均显示:第4周表达轻度升高,从第8周开始表达明显增高,CD44分子表达水平均高于对照组(P<0.05),并随着时间推移持续升高。免疫组化染色结果表明,CD44分子抗原定位于细胞膜上,诱癌组第4周开始出现弱阳性,第20周出现强阳性。结论 CD44分子在诱癌过程中的差异表达,说明其对肝癌的发生发展有明显的调控作用,但其调控机制有待进一步研究。
