摘要
目的探讨苏木的甲醇、氯仿和水提取物对甲氧西林耐药金黄色葡萄球菌(MRSA)抗菌活性,分离苏木抗菌活性成分。方法提取和制备甲醇、氯仿和水提取物,以液体试管二倍稀释法最低抑菌浓度(MIC)值比较不同提取液抗MRSA能力;高效液相色谱法分离苏木醇提物组分,M-H平皿法筛选测定各组分抑菌圈直径,液体试管二倍稀释法检测MIC值,选择最小MIC值组分鉴定,以NIH小鼠考察该组分安全性。结果苏木甲醇提液对MRSA、甲氧西林敏感金黄色葡萄球菌(MSSA)的MIC分比为200、400μg/mL(P<0.05);苏木醇提取物分离出16种组分,M-H平皿筛选组分抑菌圈直径最大为14.85mm,该组分MIC为12.5μg/mL(P<0.05),鉴定为巴西苏木素,安全实验未见不良反应。结论苏木醇提取物具有较好的抗MRSA活性;巴西苏木素抗菌活性强且无不良反应,为研发新型抗MRSA药物奠定了基础。
Objective To investigate antimicrobial activity on Caesalpinia sappan and active component against methicillin-resistant Staphylococcus aureus(MRSA) for screening drug from the traditional Chinese medicine.Methods Caesalpinia sappan physic liquor was prepared by chloroform,methanol and aqueous.They were showed antimicrobial activity against MRSA by the checkerboard dilution method and subdivided by high performance liquid chromatography.The component was used to explore antimicrobial activity against MRSA and virulence by animal experiment.Results Methanol extract of Caesalpinia sappan demonstrated a higher inhibitory activity than chloroform and aqueous extracts(P0.05).It was 200 μg/mL of methanol extract against MRSA,400 μg/mL of aqueous and 800 μg/mL of chloroform for the minimal inhibitory concentrations(MIC).Brazilin was the best powerful in the active component.It was 14.85 mm and 12.5 μg/mL to Brazilin against MRSA(P0.05).Brazilin was virulence to NIH mouse by injection.Conclusion The methanol extract of Caesalpinia sappan could have antimicrobial activity against MRSA.Brazilin might be powerful and virulence.
