摘要
目的:探讨缺氧对人肝癌HepG2细胞中缺氧诱导因子-1α(HIF-1α)及增殖细胞核抗原(PCNA)表达的影响。方法:体外培养HepG2细胞,通过CoCl2化学模拟缺氧作用,采用实时荧光定量聚合酶链式反应实验(Real-time fluorescent quantitative polymerase chain reaction,PCR)检测基因HIF-1α、PCNA mRNA水平变化,蛋白质印迹(Western Blot,WB)检测HIF1-1α、PCNA蛋白表达水平的变化,细胞化学染色检测HIF-1α、PCNA蛋白表达变化。结果:CoCl2导致的化学缺氧可诱导人肝癌HepG2细胞HIF-1α、PCNA的上调表达。结论:缺氧可提高HIF-1α的表达,可能促进肝癌细胞的进一步增殖。
Objective : To explore the effects of hypoxia on the expressions of HIF-1 α and PCNA in hepatocellular carcinoma cell HepG2. Methods : Hypoxia was imitated by adding CoCl2 in vitro. The expression level of mRNA was studied by Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR). And we used Western Blotting to explore the variations of expression in protein level. The pvotein level in vivo was also studied by cytochemical staining. Results : Higher expression of HIF-1α and PCNA in mlKNA and protein level was induced by hypoxia imitated by adding CoCl2 both in vitro and in vivo. Conclusions : Hypoxia can upregulate the expression of HIF-1 α, which may promote the proliferation of hepatocellular carcinoma cell HepG2.
