奥沙利铂体外对人视网膜母细胞瘤Y79细胞增殖及凋亡的影响 被引量:3

The effect of oxaliplatin on the proliferation and apoptosis of human retinoblastoma cells in vitro

摘要 目的探讨奥沙利铂对人视网膜母细胞瘤细胞增殖及凋亡的作用,以期为人视网膜母细胞瘤的临床治疗提供基础。方法噻唑蓝(MTT)法检测不同浓度(0、0.125、0.25、0.5、1、2μmol/L)奥沙利铂对人视网膜母细胞瘤Y79细胞的增殖抑制作用;碘化丙啶(PI)单染流式细胞仪检测奥沙利铂对人视网膜母细胞瘤细胞凋亡的影响;半胱氨酸天冬氨酸蛋白酶3(Caspase-3)活性检测试剂盒检测Caspase-3的活性变化。结果不同浓度奥沙利铂对人视网膜母细胞瘤细胞具有显著的增殖抑制作用,0.25μmol/L奥沙利铂处理视网膜母细胞的24、48、72 h后细胞的存活率分别为(83.59±0.76)%、(77.30±1.24)%、(61.41±1.39)%(P<0.05);0.25μmol/L奥沙利铂诱导Y79细胞24 h凋亡率为(36.23±0.25)%(P<0.05)。奥沙利铂增强人视网膜母细胞瘤Y79细胞中Caspase-3的活性,活性倍数为(5.76±0.25)%(P<0.01)。结论奥沙利铂具有抑制人视网膜母细胞瘤Y79细胞增殖的作用,并诱导其凋亡的作用,其机制可能与其激活Caspase-3有关。 Objective To study the effects of oxaliplatin on the proliferation and apoptosis of human retinoblastomacells,and to provide a new approach for clinical treatment of retinoblastoma. Methods MTF method was used to detect the influence of cell proliferation inhibition of oxaliplatin ; the cell apoptosis was analyzed using flow cytome- try with propidium iodide staining. Caspase-3 active detection kit was used to detect drug effect within 24h for Caspase-3 activity change. Results The different concentrations of oxaliplatin had significant proliferation inhibi- tion of retinoblastoma cells Y79. After 0.25 μmol/L oxaliplatin was administered to retinoblastoma cells Y79 for 24, 48, 72 h, the cell survival rate was (83.59 ± 0. 76) %, (77.30 ± 1.24) % and (61.41 ± 1.39) % respective- ly(P 〈0. 05). After 0. 25 μmol/L oxaliplatin was administered to human retinoblastoma cells Y79 for 24 h, the apoptosis rate was (36. 23± 0.25 ) % ( P 〈 0. 05 ), higher than the control. Oxaliplatin could enhance Caspase-3 activity in retinoblastoma cells Y79, whose activity multiple was (5.76 ± 0.25 ) % ( P 〈 0.01 ). Conclusion Ox- aliplatin has inhibitory effect on proliferation of human retinoblastoma cells Y79 and enhances the process of apopto- sis of human retinoblastoma cells Y79, the mechanism of which may relate to activation of Caspase-3.
