抗凝剂在改良MAIPA法中对ITP患者血小板特异性自身抗体(GPⅡbⅢa和GPⅠbα)检测的影响 被引量:5

摘要 探究二价阳离子络合剂的抗凝剂在改良单克隆抗体俘获血小板抗原(MAIPA)法检测免疫性血小板减少症(ITP)患者血小板特异性自身抗体中的影响。分别留取ED-TA-K2和肝素钠抗凝的162例ITP患者标本,采用改良MAI-PA法检测血小板特异性自身膜糖蛋白IIbIIIa(GPIIbIIIa)和膜糖蛋白Ibα(GPIbα)。抗GPIIbIIIa抗体:单用EDTA-K2抗凝剂检出阳性77例,单用肝素钠抗凝剂检出阳性78例,双重检测阳性102例;抗GPIbα抗体:单用EDTA-K2抗凝剂检出阳性53例,单用肝素钠抗凝剂检出阳性51例,双重检测阳性72例。单一抗凝剂与双重抗凝剂比较差异有统计学意义(P<0.05)。二价阳离子络合剂的抗凝剂在改良MAIPA法检测ITP患者血小板特异性自身抗体中有一定影响,采用两种抗凝剂双重检测可提高自身抗体的检出率。 To investigate the influence of differet anticoagulant by modified monoclonal antibody immobilization of platelet antigen assay (MAIPA), on immune thrombocytopenia (ITP) platelet specific antibodies glycoprotein IIbI- IIα (GPIIbIIIa) and glycoprotein Ibct (GPIbct). Samples from ethylene diamine tetraacetie acid dipotassium (ED- TA-K2) and heparin anticoagulation patients were obtained. 162 ITP samples were detected by modified MAIPA platelet specific autoantibodies (GPIIb Ⅲ a and GPIbα ). Group of patients were detected. Single anticoagulants group and dual anticoagulants group differences are statistically significant in detection of GPIIbIIIa and GPIbα(P 〈 0.05 ). There is significant effect in detection of different anticoagulant detected ITP platelet specific antibodies on modified MAIPA. Adopting both anticoagulants to perform combined detection can improve ITP platelet specific antibodies.
