硒对培养大鼠胎肝细胞核酸及蛋白质合成的影响

摘要 本文对比观察了硒对~3H-TdR,^(14)C-UR参入培养大鼠胎肝细胞核酸量的影响,同时还测定了培养基中白蛋白及甲胎蛋白的含量。实验结果表明,细胞培养基中硒浓度在5.7~7.6μg/L范围内使~3H-TdR,^(14)C-UR的参入量,白蛋白及甲胎蛋白的含量均明显高于不加硒组。提示硒在这一浓度范围内促进肝细胞DNA,RNA的更新和蛋白质的合成;并说明肝细胞培养的最适硒浓度为5.7~7.6μg/L。 The influnce of selenium on the syntheses of protein, DNA and RNA in the culture of feto-rat liver cells was studied by adding different amount of selenium into cell culture medium and investigating the 3H-TdR, 14C+UR incorporation into DNA and RNA, and measuring the contents of albumin and AFP (alpha-fetoprotein) in the culture medium. Results showed that the contents of albumin and AFP, the amount of 3H-TdR and 14C-UR incorporation were all much higher in cells cultivated with an adding of 5.7 - 7.6μg / L Se in the culture medium than those cultivated without Se in the culture medium, indicating that Se-sup-plement at a level of 5.7-7.6μg / L in the culture medium can improve the syntheses of protein, DNA and RNA in the feto-rat liver cells.
