摘要
目的观察青天葵(NFS)提取物对脂多糖(LPS)诱导小鼠巨噬细胞RAW264.7释放一氧化氮(NO)的影响,并探讨其抗炎作用机理。方法建立LPS诱导RAW264.7的炎症反应模型,MTT法测定NFS对RAW264.7细胞生长增殖的影响,Griess法测定NO释放量,ELISA法检测细胞上清液中PGE2、IL-6、TNF-α的含量。结果 NFS能显著降低LPS诱导的RAW264.7细胞中NO、IL-6、TNF-α的含量,并呈剂量依赖性(P<0.05或P<0.01),但对PGE2的含量无明显影响(P﹥0.05)。结论 NFS可能通过抑制LPS诱导的RAW264.7中IL-6、TNF-α的含量增加,从而产生抗炎作用。
Objective To study the Anti-inflammatory effect of the extractive from Nervilia fordii(Hance) Schltr. MethodsRAW264.7 macrophage line in mice was induced by LPS to set up the inflammatory model.Effect of different concentration of the extractive on the proliferation of RAW264.7 cells were measured by MTT,Nitric oxide(NO) production was examined by Griess reaction,the secretion of PGE2,IL-6,TNF-α in the supernatant was determined by ELISA. ResultsThe contents of NO,IL-6,TNF-α were decreased significantly in all groups,the PGE2 was no obvious influence. ConclusionThese results indicated that the extractive from Nervilia fordii(Hance) Schltr might effectively block inflammation by down-regulating production of NO,IL-6,TNF-α.
