摘要
目的研究红景天对顺铂(CDDP)所致小鼠肾损伤的防护作用,并探讨其可能的机理。方法将50只小鼠按体质量随机分5组,分别为对照组、CDDP组、低剂量红景天+CDDP组;中剂量红景天+CDDP组;高剂量红景天+CDDP组。红景天连续给予小鼠灌胃12 d,第9天时腹腔注射CDDP(20 mg/kg)。注射CDDP后物第3天采血处死动物,测定血清尿素氮(BUN)、肌酐(CRE)、肾脏系数,肾皮质蛋白、GSH含量及SOD活性。结果低剂量红景天能降低顺铂所致的小鼠血清BUN、CRE含量的升高,明显改善顺铂所致的肾组织GSH含量、SOD酶活力的变化,从而减轻CDDP的肾毒性。结论低剂量红景天经口给予能防护CDDP所致的小鼠肾损伤,其机制可能与其抗氧化作用和清除自由基活性有密切关系。
Objective To investigate the protective effects of Rhodiola on cisplatin(CDDP)-induced nephrotoxicity in mice and its possible mechanism. Methods50 male mice randomly were divided with avoirdupois into five groups:control,CDDP,low concentration Rhodiola+CDDP,middle concentration Rhodiola+CDDP and high concentration Rhodiola+CDDP.Rhodiola continuously gove mouse 12d and ip CDDP(20 mg/kg)in 9th.After CDDP treatment of animal 3 days the animals were sacrificed,and the BUN,CRE,Kidney coefficient,content of renal protein,GSH and SOD vitality. ResultsLow concentration Rhodiola can reduce the cisplatin-induced BUN and CRE elevation in serum,and improve the renal tissue GSH content and SOD activity changes,which were significant.Cisplatin-induced nephrotoxicity was alleriated. ConclusionLow concentration Rhodiola given by mouth can protect the cisplatin-induced nephrotoxicity.Antioxidation and scavenging of free radicals underlies the mechanism.
