三七总皂苷对肺纤维化小鼠组织蛋白酶B表达的影响 被引量:5

摘要 目的探讨三七总皂苷(Panax Notoginseng Saponins,PNS)对肺纤维化小鼠肺组织中组织蛋白酶B(cathepsinB,CatB)表达的影响。方法 C57BL/6雄性小鼠90只,分为假手术组,模型组和PNS组,每组30只。除假手术组外其余各组小鼠气管内一次性滴注盐酸博莱霉素(Bleomycin,BLM),假手术组小鼠气管内一次性滴注等体积的生理盐水。造模后第2天开始给药,持续到处死动物的前1天。假手术组、模型组分别灌服等体积的生理盐水。各组动物于7,14,28 d随机处死10只,取肺组织进行HE和Mallory染色;免疫组织化学法和Western-blot法检测CatB的表达情况。结果与模型组比较,PNS组肺泡炎和纤维化程度均减轻,CatB表达显著降低(P<0.05)。结论 PNS能明显减轻BLM所致的小鼠肺泡炎及纤维化程度,可能与其抑制肺组织中CatB的过度表达有关。 Objective To investigate the effects of PNS on the expression of connective tissue cathepsinB in mice pulmonary fibrosis. MethodsC57BL/6 male mice were 90,divided into sham operation group,model group and PNS group,n=30.In addition to the sham operation group,the remaining mice a single intratracheal instillation of bleomycin hydrochloride,sham operation group were a single intratracheal instillation with saline.2 days after modeling,starting dose,until the day before the animals were killed.sham operation group,model group were given equal volume of saline.All the animals at 7,14,28 d were killed 10 lungs were taken for HE and Mallory staining,Immunohistochemistry and Western-blot CatB. ResultsThe degree of alveolitis and pulmonary fibrosis in PNS group was improved compared with that of model group.PNS could reduce CatB(P&lt;0.05). ConclusionThe effects of PNS on mice pulmonary fibrosis may be correlated with inhibition of CatB over-expression in the lung tissue.
