摘要
目的:测定月季不同部位(花、叶、根)中槲皮素的含量,比较含量差异。方法:样品经甲醇-盐酸混合溶液提取水解后,采用高效液相色谱法,色谱柱为Zorbax SB-C18柱(150 mm×4.6 mm,5μm);流动相为乙腈-水(0.2%磷酸)(35∶65);流速为1.0 ml/min;检测波长为367 nm。结果:槲皮素在5.04μg/ml~50.4μg/ml范围内与峰面积具有良好的线性关系,回归方程为A=91731C-288917(r=0.9998),平均回收率99.5%,RSD=2.1%。槲皮素在不同部位的含量分布为花>叶>根。结论:该方法准确可靠,重复性好,可为月季的开发利用及质量控制提供依据。
Objective:To determine and compare the contents of quercetin in different parts(flowers,leaves and roots)of Rosa chinensis.Methods: Samples were extracted with HCl-methanol mixed solution and separated on Zorbax SB-C18 column(150 mm ×4.6 mm,5 μm).The mobile phase consisted of acetonitril-water(0.2% phosphoric acid)(35∶65) at a flow rate of 1.0 ml/min.The detection wavelength was set at 367 nm.Results: The calibration curve showed good linearity in the range of 5.04 μg/ml~50.4 μg/ml.The regression equation was A= 91731C-288917(r = 0.9998).The average recovery was 99.5% with RSD of 2.1%.The contents of quercetin in different parts of Rosa Chinensis were as follows: flowers>leaves>roots.Conclusion: This method is simple and accurate.The experimental results are reproducible.It can be used for exploitation and quality control of Rosa Chinensis Jacq.
