摘要
目的:建立食品中9种人工合成色素(新红、苋菜红、胭脂红、诱惑红、赤藓红、柠檬黄、日落黄、靛蓝、亮蓝)同时测定的高效液相色谱法。方法:饮料、配制酒样品加温除去二氧化碳或乙醇,糖制品加水加温溶解,肉制品、乳制品等高脂肪高蛋白食品先用石油醚脱脂,再加4 mol/L尿素+甲醇(1+1)震荡提取,离心,取上清液,重复直至上清液无色,低温旋转蒸发至近干,最后加水定容后测定。色谱条件:色谱柱Cnwsil C18柱,梯度洗脱,可变紫外检测波长程序480 nm~600 nm。结果:本法线性关系良好(r=0.9996~0.9999),在不同食品中3个浓度添加水平平均回收率为77.6%~111.7%,相对标准偏差为0.6%~4.2%,最低检出浓度为0.100~0.500 mg/kg。结论:本法简便实用、准确快速,灵敏度高,特异性强,适用范围广,可满足食品中多种色素同时分析要求。
Objective:To develop a high performance liquid chromatographic method for simultaneous determination of 9 synthetic pigments,including new red,amaranth,ponceau,allura red,erythrosine,tartrazine,sunset yellow,indigotine,brilliant blue in foods.Methods: Beverages,wine samples were heated to remove carbon dioxide or ethanol,sugar products were dissolved in water by heating;meat products and dairy products with high fat and high protein were degreased with petroleum ether,then the pigments in sample solutions were extracted with 4mol/L urea and methanol(1+1),and centrifuged,then the supernatant was taken out and the procedure was repeated until the supernatant became colorless.The supernatant was rotary evaporated to nearly dry,redissolved in water and finally separated on column Cnwsil C18 with gradient elution and detected at 480 ~600nm.Results: The method showed good linearity with r =0.9996 ~0.9999.The average recoveries were 77.6% ~ 111.7% with RSDs of 0.6%~4.2%,the minimum detectable contents were 0.100mg/kg ~0.500mg/kg.Conclusion: The method is simple and practical,accurate and rapid,high sensitive and specific,it can be used for the determination of multiple synthetic pigments in foods.
