针康法对局灶性脑缺血大鼠运动功能及缺血区周围内皮素受体-β表达的影响 被引量:7

Effects of Cluster Needling of Scalp Acupuncture Combined with Rehabilitation on Motor Function and Expression of Endothelin - β(ET - β) after Focal Cerebral Ischemia

摘要 目的:观察针康法对局灶性脑缺血大鼠运动功能及缺血区周围内皮素受体-β表达的影响。方法:选取清洁级SD雄性大鼠,按随机数字表法分成5组,假手术组、模型组、针刺组、康复组以及针康组,按时间点每组又分为3天、7天、14天、21天共4个亚组。采用线栓法制备脑梗死动物模型,造模后24 h分别进行相应干预。针刺组仅采用头穴丛刺,康复组仅进行康复训练,针康组采取头穴丛刺结合康复训练,采用行为学评分法分析大鼠运动功能,并应用免疫组化法观察各组不同时间点缺血灶周围皮质ET-β受体表达。结果:针康组术后14天、21天行为学评分低于针刺组、康复组,且针康组明显低于模型组(P<0.05);造模后大鼠ET-β受体阳性细胞数增加,3天和7天达到高峰,随后逐渐减低;3天、7天和14天时,针康组ET-β受体阳性细胞的表达水平低于针刺组、康复组,明显低于模型组,差异具有统计学意义(P<0.05)。结论:针康法可促进局灶性脑缺血大鼠运动功能的恢复,其机制可能是与减少缺血区周围ET-β受体的表达有关。 Objective: To explore the effects of cluster needling of scalp acupuncture combined with rehabilita- tion on motor function and expression of endothelin - β ( ET - β) in rats after focal cerebral ischemia. Methods: Select clean Sprague - Dawley (SD) male rats and they were randomly divided into five groups : sham operation group, model group, rehabilitation group, acupuncture group, and acupuncture with rehabilitation group, each group was divided into four subgroups by 3 d,7 d, 14 d and 21 d. Application Longa line bolt for preparation of rat middle cerebral artery occlusion model. Sham operation group was from postoperative spontaneous recovery. Head hole cluster needling group was with per head hole cluster needling therapy, acupuncture group was only with head acupuncture, rehabilitation group was only performed treadmill training, needle Kang group adopted the head acupuncture combined with treadmill training, and the application behavior scoring analysis was used for rat motor function recovery. Take the brain using immunohistochemistry observation each time point cerebral infarction surrounding white ET-β expression. Results: Needle Kang group after 14d, 21d behavior score was below the head acupuncture group and rehabilitation training group, and it was significantly lower than model group, the difference was statistically significant (P 〈0.05) ;needle Kang group, head acupuncture group, re- habilitation training group, the two rat ET - receptor positive cells in the cerebral ischemia after modeling be- gan to increase, respectively, in 3d and 7d it reached the peak , then gradually decreased; cerebral isehemia at different time points after needle Kang group, ET - β expression levels were lower than that in the head acu- puncture group, and it rehabilitation training group, and it was significantly lower than model group ( P 〈 0.05 ). Conclusion : The cluster needling of scalp acupuncture combined with rehabilitation can promote recovery of mo- tor function in rats after focal cerebral ischemia, which may be associated with reduced expression of ET - β .
