Determination of chlorogenic acid and liriodendrin in Eucommiae Cortex by HPLC-ELSD

摘要 目的建立HPLC-ELSD法同时测定杜仲中绿原酸和鹅掌楸苷的方法。方法奥泰C18柱(250mm×4.6mm,5μm);流动相:乙腈-0.2%醋酸水,梯度洗脱;体积流量:0.9mL/min;柱温:35℃;漂移管105℃,气体流量:2.5L/min。结果绿原酸和鹅掌楸苷分别在0.0214~4.2800μg和0.0206~4.1200μg呈良好的线性关系;绿原酸和鹅掌楸苷紫的平均回收率分别为98.57%、103.37%,RSD值分别为2.23%、2.56%。结论本法简便、准确,可用于杜仲中绿原酸和鹅掌楸苷的测定。 Objective To establish the method for the simultaneous determination of chlorogenic acid and liriodendrin in Eucommiae Cortex by HPLC-ELSD.Methods Alltima C18 column(250 mm×4.6 mm,5 μm)was used.The mobile phase consisted of acetonitrile-0.2% acetic acid solution and the gradient elution was carried out.The flow rate was 0.9 mL/min.The temperature of column and drift tube was set at 35 ℃ and 105 ℃,respectively.The flow rate of gas maintained at 2.5 L/min.Results The linear ranges of chlorogenic acid and liriodendrin were 0.021 4 — 4.280 0 and 0.020 6 — 4.120 0 μg,respectively.The average recovery rates of chlorogenic acid and liriodendrin were 98.57%(RSD=2.23%)and 103.37%(RSD=2.56%).Conclusion The method is simple,convenient,and accurate,and could be used for the determination of chlorogenic acid and liriodendrin in Eucommiae Cortex.
