高效液相色谱法对绵萆薢质量控制的研究

QUALITY CONTROL OF DIASCOREA SEPTEMLOBA THUNB BY HPLC

目的优化测定绵萆薢中薯蓣皂苷元含量的高效液相色谱法,为评价绵萆薢药材质量提供可行的方法。方法色谱柱为symmetryshield TMRP18(150.0mm×3.9mm,5μm),以乙腈-水(80∶20)为流动相,流量1.2mL/min,检测波长203nm,柱温为常温。结果薯蓣皂苷元进样量在0.019 8～0.505 0g/L范围内线性关系良好(r=0.999 5),平均回收率为98.23%。结论本方法操作简便、准确,可用于绵萆薢的质量控制。

Objective To optimize an HPLC method for determination of Diosgenin in Dioscorea septemloba Thunb and provide a basis for assessing the quality of this drug. Methods The column was symmetryshield TMRP18 （150.0 mm×3.9mm,5μm） ;The mobile phase consisted of acetonitrile＇water （80 ： 20） at a flow rate of 1.2 mL/min; The determination wave length was set at 203 nm and the column temperature was room temperature. Results The linear range of Diosgenin was 0.019 8-- 0.505 0 g/L （r=0.999 5）, the average recovery was 98.23%. Conclusion The established method is simple, convenient and ac- curate, which can be utilized for quality control of Dioscorea septemloba Thunb.
