The Effect of VEGF on iNOS Protein Expression in HaCaT Cells and Its Relationship with the Cell Proliferation

摘要 目的探讨VEGF对HaCaT细胞诱生型一氧化氮合酶蛋白表达的影响。方法采用MTT法进行细胞增殖实验;Westernblot检测iNOS蛋白表达;Griess法检测NO的含量。结果 VEGF能促进HaCaT细胞增殖;VEGF作用后HaCaT细胞iNOS蛋白的表达上调,HaCaT细胞上清中NO含量增加;加入L-NAME后,VEGF促进HaCaT细胞增殖作用降低。结论 VEGF能促进HaCaT细胞诱生型一氧化氮合酶表达,且后者与HaCaT细胞增殖相关。 Objective To investigate the effect of ~EGF on iNOS protein expression in HaCaT cells and the relationship with the cell proliferation. Methods The protein of iNOS in HaCaT cells was measured by Western blot; NO levels in HaCaT cellular supernatant were measured by Griess method; the effect of proliferation on HaCaT cells treated by VEGF and nitric oxide synthase inhibitor ( L-NAME ) were measured by MTF. Results Within the selected concentrations (5ng/mL, 10ng/mL, 25ng/mL, 50 ng/mL), VEGF could promote the proliferation of HaCaT cells. The protein of iNOS and NO levels in HaCaT cells stimulated by VEGF was significantly increased. L-NAME could inhibit the proliferation of HaCaT cells stimulated by VEGF (10ng/mL)at 48h and in a dose-dependent manner. Conclusion VEGF could enhance iNOS protein expression in HaCaT cells. Meanwhile, the expression of iNOS protein might be closely correlated with the proliferation of HaCaT cells stimulated by VEGF.
