摘要
对来自江西、广东、上海三省市的82个副猪嗜血杆菌野外分离株进行SDS-PAGE分析,采用强毒力野外分离株的多克隆抗血清,通过Western blotting对副猪嗜血杆菌OMP和全细胞蛋白的免疫原性进行研究,寻找具有共同抗原决定簇的免疫原性蛋白。结果表明免疫血清对不同来源的分离株具有良好的交叉免疫反应,患病猪的分离株OMP和全细胞蛋白具有较强的免疫原性,健康猪的分离株OMP免疫原性较全细胞蛋白免疫原性弱。不同菌株的共同免疫原性蛋白为OMP中相对分子质量为37 000~40 000、28 000~30 000的蛋白。
The out membrane protein (OMP) in 82 isolations of Haemophilus Parasuis from Jian gxi,Guangdong and Shanghai were determined by SDSPAGE. The highly virulence isolates were used for preparing polyclonal antiserum,and western blotting were used for researching the immu nogenicity of OMP and wholecell proteins, and searching for the antigen with common antigenic determinants. The results showed that the 5 kinds of polyclonal antiserum could reacted remarked ly to the OMP of homologous isolates,and the relative molecular weights of the main immune re action proteins were 28 000-30 000 and 37 000-40 000. The antiserum could also reacted obvi ously to various component of wholecell proteins as well. Five kinds of immune serum showed a good effect in crossimmune against strains of different clinical sources. Both OMP and wholecell proteins from disease pigs showed good immunogenicity. OMP of isolates from healthy pigs indica ted weaker immunogenicity than wholecell proteins. However, the immunogenicity of isolates ()MP in disease pigs were stronger than in healthy pigs. It was found that the typical immunoge nicity proteins among different strains were relative weight between 37 000 and 40 000,28 000 and 30 000.
